Bioactive peptides from marine sources: pharmacological properties and isolation procedures OceanDocs

Marine organisms represent a valuable source of new compounds. The biodiversity of the marine environment and the associated chemical diversity constitute a practically unlimited resource of new active substances in the field of the development of bioactive products. In this paper, the molecular diversity of different marine peptides is described as well as information about their biological properties and mechanisms of action is provided. Moreover, a short review about isolation procedures of selected bioactive marine peptides is offered. Novel peptides from sponges, ascidians, mollusks, sea anemones and seaweeds are presented in association with their pharmacological properties and obtainment methods.

Despite the fact that the involvement of environmental factors in the physiology of the japanese oyster C. gigas is fully described, regulatory peptides are not known. In this study, we focused on peptidergic control of egg-laying. In order to characterize peptides involved in the regulation of egg-laying in C. gigas, the cuttlefish S. officinalis was used as a model. Firstly, regulatory peptides were investigated in the cuttlefish by peptidomic analysis. This approach led to the identification of eleven new ovarian regulatory peptides. The production of fish larvae by hatchery shows an intensive increase for several species, but the specific nutritional requirements of larvae were poorly known until some years. We have conducted several experiments aimed to understand the specificities in larvae digestion, and to formulate a compound diet taking into account the differences compared to juveniles. The chosen species was the European sea bass, Dicentrarchus labrax, and the data were validated on different species. Marine fish larvae undergo major functional and morphological changes during the developmental stages and several factors can interfere with the normal development of larvae and affect fry quality. Skeletal malformations, such as spinal malformation-scoliosis, lordosis, coiled vertebral column-, missing or additional fin rays, bending opercle or jaw malformations, are frequently observed in hatchery-reared larvae. The effects of dietary marine protein hydrolysates on the development of sea bass larvae, Dicentrarchus labrax, and associated microbiota ArchiMer

Protein hydrolysate is an essential component of dry starter diets for fish larvae, as promoting healthy development. Peptides are also suitable substrates for many intestinal microbes. Five experimental diets were compared to a control diet (CONT) supplemented with a commercial fish protein hydrolysate. Each diet contained one marine protein hydrolysate, which differed by the proportion of di- and tri-peptides, and by raw materials. Two diets (HYD4 and HYD5) stimulated larval growth compared with CONT. Two other diets (HYD1 and HYD2) yielded inferior growth. The fact that Alzheimer's beta amyloid (A&#946;) peptides forms cation channels in lipid bilayers was discovered during the course of our experiments in the laboratory of "Guayo" Rojas at NIH in Bethesda, Maryland (USA). Recently, we found that the A&#946; ion channel could be blocked selectively with small peptides that copy the amino acid sequence of the predicted mouth region of the A&#946; channel pore. We now have searched for the essential amino acid residues required for this blocking effect by mutations. We found that the ability of peptides to block A&#946; channel activity could be lost by replacement of histidines 13 and 14 by alanine or lysine. Production of Hydrolysate of Okara Protein Concentrate with High Antioxidant Capacity and Aglycone Isoflavone Content BABT

Abstract This work aimed to evaluate the enzymatic hydrolysis of okara protein concentrate with respect to degree of hydrolysis (DH) in order to obtain a protein hydrolysate with high antioxidant capacity and aglycones isoflavone content. A central composite rotatable design was carried out to evaluate the influence of temperature (40 to 70°C), enzyme:substrate ratio (0.5 to 5.0%, g/100g protein) and pH (7.0 to 9.0) on DH. The optimal condition was 55°C, pH 9 and enzyme:substrate ratio of 5.0%, resulting a DH value of 35.5%. Endogenous vasopressin and the central control of heart rate during dynamic exercise BJMBR

The present article contains a brief review on the role of vasopressinergic projections to the nucleus tractus solitarii in the genesis of reflex bradycardia and in the modulation of heart rate control during exercise. The effects of vasopressin on exercise tachycardia are discussed on the basis of both the endogenous peptide content changes and the heart rate response changes observed during running in sedentary and trained rats. Dynamic exercise caused a specific vasopressin content increase in dorsal and ventral brainstem areas. Action of a pancreatin and an Aspergillus oryzae protease on whey proteins: correlation among the methods of analysis of the enzymatic hydrolysates BABT

The objectives of this study were to hydrolyze whey proteins using a pancreatin and an Aspergillus oryzae protease; to evaluate the degree of hydrolysis (DH) and the peptide profile; and to establish the correlations among the analytical methods. Ten hydrolysates were prepared at different reaction times and the highest DH was obtained by the protein content method. Good correlations (r > 0.87) between the methods of formaldehyde and orthophthalaldehyde (OPA), formaldehyde and osmometry as well as osmometry and OPA were observed using pancreatin. Similar results were obtained between OPA and soluble protein content for the A. oryzae protease. The main objective of this study is the valorisation of by-products coming from processed tuna (Thunnus albacares). Therefore, enzymatic hydrolysis was selected, using an industrial large spectrum protease (Protamex).
